Effect of Preservation Solution Containg FDP on Ultrastructural Changes During Cardiac Procurement for Transplantation in Rat


Abstract
A rat heart transplantation model is used in an experimentalstudy whethermyocardial protection is enhanced when FDP is added to preservation solution (St. Thomas hospital solution).
The effect of cardiac preservation solution on cardiac function and ultra-structure was studied in intraperitoneal heterotopic cardiac transplantation model of Sprague-Dawley rat. Preservation solution was cold (0-4¡É) St. Thomas hospital
solution.
The degree of admage of donor myocardium during preservation was quantified by means of electron-microscopic (EM) examination.
In control group (N=10), donor hearts were preserved in cold St. Thomas' hospital solution for 4 hours. In experimental group (N=10). Hearts were preserved in cold St. Thomas hospital solution containing FDP for 4 hours prior to transplantation.
EM studies of the left ventricle of transplanted heart revealed heart revealed that Flameng's scores for mitochondrial damage were significantly lower in experimental group than control (cotrol : 1.82, experimental : 1.16) (p<0.01). Cardiac
beating
score was not significant (p>0.05).
In conclusion, it is suggested that adding FDP to preservation solution improve myocardial protection for transplantation.
